What Squalene Is
Squalene is a type of compound called a triterpene. The simplest way to think about it is that it belongs to the same broad chemical family as cholesterol and many of the hormones your body makes. In fact, your liver produces squalene as a direct step in the process of making cholesterol. Every human body makes it continuously.
Your skin naturally concentrates squalene in the sebaceous glands, which produce the oil that coats and protects the skin. This natural oil, known as sebum, contains roughly 10% to 12% squalene, making it one of its most abundant components. On the skin’s surface, squalene helps support the skin barrier, reduce moisture loss, and act as an antioxidant by neutralizing free radicals before they can damage skin cells.

Why Squalene Declines as You Age

In populations that do not eat much olive oil, estimated daily squalene intake is below 30 mg. In Mediterranean populations where EVOO is the primary cooking fat, daily intake is estimated at 200 to 400 mg. This gap is large enough that researchers studying Mediterranean diet populations often measure squalene intake separately as its own variable.

How to Apply Four Hour Olive Oil to Your Skin

Skin and oxidative protection

Squalene vs. Squalane: Not the Same Compound
Squalene and squalane appear on ingredient labels with names similar enough to cause genuine confusion. Although they are closely related, they are not the same compound and behave differently in practice.
Squalene is the natural form found in EVOO and human sebum. It contains six double bonds that make it biologically reactive and allow it to function as an antioxidant at the skin’s surface. However, those same double bonds make squalene less stable in cosmetic formulations because unsaturated compounds can oxidize over time when exposed to air. This is why cosmetic manufacturers generally do not use natural squalene directly.
Squalane is squalene that has been fully hydrogenated, meaning all six double bonds have been removed. The result is a more stable molecule that is much less susceptible to oxidation and has a longer shelf life. Most skincare products that list squalane use this hydrogenated form.
